1. MacroGenics乳腺癌单抗新药达到3期临床终点
日前，专注于开发创新单克隆抗体癌症疗法的临床阶段生物医药公司MacroGenics宣布，其在研单抗药物margetuximab，在治疗转移性人表皮生长因子受体2阳性（HER2+）乳腺癌的关键3期试验SOPHIA中，取得了积极结果。MacroGenics计划于2019年下半年为margetuximab提交生物制剂许可申请（BLA）。Margetuximab是MacroGenics公司研发的对抗体Fc端进行优化的HER2单克隆抗体。
2. 拜耳darolutamide组合疗法3期结果公布，延长前列腺癌无转移生存期22个月
2月14日，治疗非转移性去势抵抗性前列腺癌（nmCRPC）患者的关键性3期ARAMIS试验结果显示，在研药物darolutamide加雄激素剥夺疗法（ADT），与安慰剂加ADT相比，统计显著改善无转移生存期（MFS），将肿瘤转移或死亡风险降低59％。Darolutamide组的中位MFS为40.4个月，安慰剂组为18.4个月，中位MFS的总体差异为22个月。Darolutamide是一种在研非甾体雄激素受体拮抗剂，它能够抑制雄激素受体功能和前列腺癌细胞的生长。
3. 第一三共CSF1R抑制剂上市申请获得FDA优先审评
2月9日，第一三共（Daiichi Sankyo）公司宣布，该公司为CSF1R抑制剂pexidartinib递交的新药申请（NDA）已经被美国FDA接受，同时FDA授予这一申请优先审评资格。Pexidartinib用于治疗腱鞘巨细胞瘤（TGCT），这是一种罕见的失能性肿瘤。如果获得批准，pexidartinib将成为治疗TGCT的第一个也是唯一的获批疗法。
4. Trovagene公布onvansertib与Zytiga联合治疗前列腺癌的2期研究进展
Trovagene是一家临床阶段的肿瘤治疗公司，采用精准医学方法开发针对细胞分裂（有丝分裂）的药物，以治疗白血病，淋巴瘤和实体肿瘤癌。2月14日，Trovagene公布了其正在进行的2期研究进展，该研究评估onvansertib联合Zytiga（醋酸阿比特龙）/泼尼松治疗转移性去势抵抗性前列腺癌（mCRPC）患者的疗效和安全性。试验数据证明了onvansertib与Zytiga联合治疗的安全性和耐受性。Onvansertib是一种“first-in-class”的第三代口服和高选择性丝氨酸/苏氨酸polo样激酶1（PLK 1）抑制剂，PLK酶在多种癌症中过量表达，包括白血病，淋巴瘤和实体瘤。
